Definition | A high-performance liquid chromatograph that separates and analyses compounds present in a liquid sample. Separation results from the difference in the relative distribution ratios of the solutes between a solid stationary phase packed into a column and a liquid mobile phase. The separated compound bands which elute from the column are then measured using a detector. HPLC forces the mobile phase under high pressures which is distinguished from traditional liquid chromatography which relies on the force of gravity. The HPLC also has better separation of the components due to the smaller particle size of the column packing material. HPLC instruments usually consist of a reservoir of mobile phase, a pump, an injector, a separation column and a detector. | ||
